Analysis of Genetic Diversity and Development of A Core Collection in Walnut (Juglans regia L.) Germplasm Native to China via Genotyping-by-Sequencing

Popular knowledge of the population structure and genetic diversity a plant species is essential for designing improvement strategies. The genotyping-by-sequencing (GBS) approach has been used to simplify complex genomes become popular high-throughput molecular tool selecting breeding many crop plants, including those with large genomes. This study aimed construct core collection walnut (Juglans regia) germplasm using GBS approach. A panel 87 initial genotypes, 25 landraces, 12 cultivars, 50 seedling populations, mostly native Gansu Province China, was subjected GBS. total 110,497 high-quality SNPs were identified determining distinct clusters an optimum number subpopulations. Structure analysis divided genotypes into three groups, which coincided their site year, suggesting certain degree separation in geographical origin pedigree among groups. To maximize utilization, posteriorly grouped according subgroups obtained through analysis. minimize subsample redundancy, designed set 6540 distributed across all 16 chromosomes. Finally, comprising nine (10% entire genotype set), five one landrace, assembled. Genetic indicated that uneven distribution landrace collection, could be related environmental conditions, are similar. Overall, results this establishment will facilitate future programs.
